When I have collected 100  Homestead Assaults, I moved to 38S, 48E and actived the mission "A Helpless Prospector".
But there is nothing and I can't finish the mission. Can somebody tell me what to do?

Make the mission active before you move on to  38s, 48e. Hope this helps Wink

I have tried again but the screen only shows "No actions available"
Does the reward appear in random with {Mission:Active; Homestead card>100; At 38S, 48E} all done?
Or am I have to wait for some hours at that location as the mission said?
Can anybody help me to check this mission problem?

that's the problem then.  A homestead that shouldn't have been reclaimed was.  See if it doesn't work for you now.
